
The Italian international picked up the injury in Roma's 2-1 win over Atalanta on the weekend.
"Totti will not be part of the squad because he hurt his ankle on Saturday," Roma's coach Luciano Spalletti was quoted as saying by Italian media on Monday.
Spalletti also revealed that midfielder Simone Perrotta is also doubtful for Tuesday's game.
"I also have doubts over Simone Perrotta, I hope he will be able to play but I cannot guarantee this will be the case," Spalletti added.
But the Roma coach said he was ready to deal with the absence of his two key-players.
"If he (Perrotta) does not recover, I have several alternatives.
"Both Vincenzo Montella and Mirko Vucinic are feeling well, although it is normal for them to not be at their best seeing as they are not playing with continuity," he added.
Roma need a draw in their final Group D game to secure passage to the knockout stage with opponents Valencia.
