The former Italy playmaker injured his knee in Friday's training session.
Totti, who made few appearances this term, has struggled to regain his fitness since undergoing an operation on the same knee in the summer.
"In these conditions, Totti cannot play," Roma coach Luciano Spalletti was quoted as saying by Calcio Italia.
"He has played three games recently and has not been able to train. This morning his knee gave him bother. He tried to accelerate, but he couldn’t do it.
"To take him with us [to Turin] would also hurt the team. We are doing everything to get him well again."
The news comes as a blow to the struggling outfit, who will be looking to end a three-match losing streak at the expense of Juventus.
Roma, who finished second in the Serie A last season, lie 16th in the league table with seven points from eight games.
