Inter Milan, Liverpool, Atletico Madrid and Sporting Lisbon are almost there, while others are aiming to join in as matchday five closes.
Bottom teams have a chance to get out with something as the challenge for the UEFA Cup spot is on as well.
Group A
The tightest group of all; with fourth-placed CFR Cluj trailing leaders Chelsea by three points ahead of the Romanians' clash with Roma.
Cluj stunned Roma at Olimpico in the opening day, but the Giallorossi have a chance to avenge their embarrassing defeat and edge closer to the last 16 with an away win.
Alberto Aquilani is fit again for Roma, Christian Panucci is a slight doubt, while Cluj are without Didi, Tony, Diego Ruiz and Gabriel Muresan.
Premiership leaders Chelsea visit Bordeaux in an attempt to seal qualification, amid the Blues' endless injury problems.
Michael Essien, Ricardo Carvalho, Juliano Belletti and Alex are all ruled out, while Deco is suspended following his red card in the 3-1 defeat at Roma.
Laurent Blanc's Bordeaux are without veteran keeper Ulrich Rame, and the inexperienced Mathieu Valverde will deputize.
Group B
Jose Mourinho's Inter are one step closer to the knockouts as they take on UEFA Cup hopefuls Panathinaikos, who were beaten 2-0 at home in the reverse fixture.
Christian Chivu and Patrick Vieira are ruled out with injuries for Inter, Panathinaikos duo Bryce Moon and Cleyton are also set to miss the game.
In Cyprus, debutants Anorthosis are poised to seal a historic qualification if they beat Werder Bremen, coupled by a win for Inter at San Siro.
Temuri Ketsbaia's men were only beaten once in the group, losing away to Inter by a single goal. Bremen are winless at the bottom.
Anorthosis miss goalkeeper Arian Beqaj and midfielders Paulo Costa (flu) and Cedric Bardon (ankle), while Bremen are without Ivorian striker Boubacar Sanogo, who has flu.
Group C
Barcelona have already qualified as they face Sporting Lisbon, who need a point to join the Catalans to the knockouts.
Captain Carles Puyol and midfielder Seydou Keita are rested for Barca, but Lionel Messi and Rafael Marquez return after missing domestic action at the weekend.
In Donetsk, Shakhtar host Swiss outsiders Basel as the Ukrainians push for a UEFA Cup spot.
Shakhtar will be without suspended Brazilian striker Brandao, and influential defender Dmytro Chygrynskyi is struggling with a thigh injury.
Group D
Liverpool and Atletico need draws from home games against Marseille and PSV Eindhoven respectively.
Rafa Benitez's side are boosted by the return of pivotal skipper Steven Gerrard, who missed the goalless draw with Fulham at the weekend with a groin injury.
Marseille stunned the Reds 2-1 in the corresponding fixture last year, but they will miss the injured Mathieu Velbuena, who scored both goals at Anfield at the time. African duo Elamine Erbate and Modeste Mbami are also ruled out.
At Vicente Calderon, Atletico entertain PSV behind closed doors as the Primera Liga side serve a UEFA stadium ban.
Spanish international Antonio Lopez misses out for Atletico, while Dirk Marcellis, Jan Kromkamp and Nordin Amrabat are ruled out for PSV.
