The Premiership big spenders are expected to bolster their team with several high-profile signings in the January transfer window.
"I have heard about the big project at City and I'm flattered by their interest," Senna told the Daily Mail.
"The Premier League is a big league and I am interested in the challenge."
However, the 32-year-old, who still have two years remaining on his Villarreal contract, said he would only leave his current club should City make a worthwhile offer.
"It is complicated leaving Villarreal because I am excited by the season and we are still involved in the league and the Champions League," he added.
"It would take an enormous proposition by City for me to leave but it is not impossible."
The Brazilian-born footballer was once close to joining the Premiership side two years ago, but the deal eventually fell through.
