"He'll play," Portuguese coach Mourinho, whose competitive debut with Real ended in a goalless draw with Mallorca in their league opener, told a news conference.
"He's in good shape and because it's a difficult match he'll play from the start."
Ronaldo sustained the injury against Mallorca and was due to be sidelined for around three weeks but he recovered quicker than expected.
His return will hand Mourinho a major boost ahead of Real's Champions League group-stage opener against Ajax Amsterdam on September 15.
Real are already without midfielders Kaka (knee) and Fernando Gago (knee) and defenders Raul Albiol (ankle) and Ezequiel Garay (knee), who are all injured.
